BIOC2101 PRINCIPLES OF BIOCHEMISRTY SCIENTIFIC REPORT ASSIGNMENT

CONGRATULATIONS!

You have been assigned ‘Mystery Enzyme A’ for your BIOC2101 Scientific Report investigation. See
below for its associated properties and experimental data that you will use for your report.

Mystery Enzyme A Data Sheet

 Isolated from: Human saliva

 Molecular weight: 14.7 kDa
 Turnover number: 0.5 s -1
 Experimental method: Firstly, ‘stopped’ enzyme assays were used to determine the variation
of reaction rate with time for ten different substrate concentrations. The resulting data was
then used to determine the initial reaction velocity for each substrate concentration.
 Experimental Data:

Data Table 1: Raw values for variation of initial reaction velocity with substrate concentration

Enzyme substrate Initial reaction velocity V0 (µmol

concentration [S] (mM) product released per second)
0.001 0.071
0.002 0.125
0.003 0.167
0.004 0.200
0.005 0.227
0.006 0.250
0.007 0.269
0.008 0.286
0.009 0.300
0.010 0.313

Use the above data to determine K M and Vmax values for this enzyme-substrate complex. (HINT: Look
at the relevant lecture notes for the course on this topic for information on how to do this).

Follow the assignment instructions (separately posted in Moodle) carefully to present and discuss
these results in your BIOC2101 Scientific Report.
