SECTION 1.8 : SAMPLING WITH A


SAMPLING DEVICE OR COIL

1. INTRODUCTION
a) Certain analyses on the product in process (contained in bulk vessels i.e. FV,
SV and BBT) may require special sampling techniques in order to ensure that
the Quality Requirements of Sampling are met.
b) Divisional sampling standards are defined in Section 3.15.
c) References: SAB Analytical MDT 1995 & 1999.

2. HEALTH AND SAFETY


2.1 HAZARDS
a) Working with glass apparatus.
b) Attaching Tygon tubing to glass tubing (apparatus).

2.2 SAFETY REQUIREMENTS


Hand towel.

2.3 PRECAUTIONS
Protect hands with a towel when inserting glass tubing into rubber or Tygon
tubing.

3. APPARATUS
3.1 EQUIPMENT REQUIRED
a) Sampling device - A. (See Appendix A)
b) Sampling coil - B. (See Appendix B)
Note: i) These devices can be made of glass or high quality stainless
steel.
ii) If glass - made up by a glass blower.
c) Glass beaker - 100 ml.
d) Beer bottle - 750, 375 or 340 ml.
e) Tygon tubing = ID 1 mm less than the OD of the glass or stainless steel
tube.
f) Screw clamp.
g) Bottle crowns.
h) Hard crowner.

3. APPARATUS (Continued)

3.2 PREPARATION REQUIRED


a) Device A: Attach Tygon tubing (about 20-30 cm) to the Inlet and Outlet tubes and
attach a screw clamp to the Tygon tubing on the Outlet.
b) Coil B: Attach Tygon tubing (about 10-20 cm) to the Inlet tube.
Note: Before attaching Tygon tubing to glass tubes, heat the end of the
Tygon tube ( 2-3 cm) in a beaker containing hot water.

3.3 MAINTENANCE
a) Sampling devices must be cleaned immediately after use.
b) Tygon tubing must be inspected frequently and replaced if worn.

a) Detergent (Laboratory grade).
b) Carbon dioxide (cylinder).
c) Crushed ice.

5. METHOD
NB: Adhere to manualised requirements in sections 3.2 and 3.15 of the
manual.

5.1 SAMPLING DEVICE A


This device is used to avoid the loss of CO 2 or formation of foam.
a) Attach the sampling device to a clean 750 ml (375 or 340 ml) beer bottle.
b) Loosen up both screw clamps.

5.1 SAMPLING DEVICE A (Continued)
c) Attach the inlet section of the Tygon tubing to the sample cock supply of
carbon dioxide (CO2).
d) Flush the beer bottle 2-3 minutes with CO 2 and close off the supply.
e) Immediately close off both screw clamps.
f) Attach the inlet of the Tygon tubing to the tank’s sample cock.
Note: Pre-flush the sample cock/main.
g) Crack open the screw clamp on the outlet of the sampling device and fully
open the screw clamp on the inlet.
h) Gently open the tank sample cock.
i) Regulate the flow rate with the outlet screw clamp such that the product fills
the bottle slowly and gently.
Note: No gas bubbles or foam must be visible.
j) Allow clear liquid to run from the outlet for about 2 minutes and close the
tank sample cock.
k) Remove the sampling device and immediately crown the bottle.
Note: i) Ensure a headspace of about 15-25 mm in the bottle prior to
crowning.
ii) Prepare and analyse the sample as per the manual method.

5.2 SAMPLING COIL B


a) Flush the sample cock and main on the tank.
b) Attach the coil, via the Tygon tubing, to the sample cock.
c) Open the sample cock slowly and allow clear liquid to flow through the coil.
Note: i) It may be necessary (as per the relevant method) to house
the coil in a beaker containing ice water (i.e. sampling for SO 2
etc.)
ii) The sample is collected ex coil as described in the respective
method.
d) Close the sample cock after sampling.

6. VERIFICATION
a) A representative and homogeneous sample.
b) Refer to Section 3.12 Verification Routines.

7. COMPETENCY TEST
No Formative Assessment Checklist (FAC) will be developed for this section. Where
relevant each method will contain the appropriate and relevant FAC requirements
necessary.

ii) Dimensions for stainless steel may be

steel may be about 2 mm smaller.
