AP NOTES – Cytology and Microscope Worksheet

I. Introduction
The human body is divided into two types of structures: those that are large enough to be seen
with the naked eye (macroscopic structures) and those that are small enough to be seen only
with the use of a microscope (microscopic structures) (Boundless, n.d.). Microscopy, which is
the investigation of microscopic structures using a microscope, an equipment used to acquire
an enlarged image of small objects, and Cytology, which is the study of the origin, structure,
and function of cells, will be studied in this topic (Biology Online Dictionary, n.d.). To fully grasp
the subject, the components, functioning, and use of a Brightfield microscope will be covered.
Through the use of a Brightfield Microscope and a brief discussion of the structural organization
of an organism and cell homeostasis, the goal of this worksheet is to gain a knowledge of how
Microscopy and Cytology function together. By the end of this, the students will be able to see
a link between all of the previously mentioned topics and the importance of understanding
them.

II. Guide Questions


a. Explain the importance of using a low power objective in initiating the use of a
microscope.
b. Provide an explanation of the use of the coarse focus knob to each objective
lens.

i. Compute for the Total Magnification
ii. Suppose that in further using the Brightfield Microscope, the next views
generated were the result of rotating the objective lenses to the next
power and then to the highest power. Compute the total magnification
for each, and explain your answer.
d. Define the structural organization that makes up an organism.
e. Explain the mechanisms that maintain cell homeostasis.
