동물세포 배양
(2) 배양 목적
• 효소, 호르몬, 백신, 항암제 등 의료용 단백질 (바이오의약품)을 생산
• 의료용 단백질은 유전자 재조합된 미생물을 이용하여 생산할 수도 있다.
• 그러나 동물세포를 이용하는 것이 제품 (최종 생성물)의 품질면에서 바람직
• 이들 생산에 필요한 최적화된 배양조건 및 세포내 신호 (signal) 기작을 연구
• 동물세포는 부착 (attachment) 상태나 부유(suspension) 상태로 배양
동물세포 배양: 고려해야 할 점
• 부착세포의 scale-up
• 병 속에 있는 배양배지를 회전시킴
• 대량생산에 필요
현탁 배양기: 바이오리액터
• Batch (회분식): 일정량의 배양배지 이용
• Fed-batch (유가식): 농축된 배지를 추가적으로 공급
• Continuous (연속식): 새 배지를 연속적으로 리액터에 공급하고 오래된 배지를 연
속적으로 제거

동물세포 배양에 필요한 기기: CO2 incubator
• 무균조작을 위한 작업대
• HEPA filter 및 송풍기를 사용하여 무균공기를 항상 내부로 보냄
• 내부에는 형광등과 자외선 살균등이 설치되어 있어야 하며, 사용하지 않을 때에는 항
상 살균등을 켜놓아야 한다.
• 사용한 후에는 작업대를 70% ethanol로 닦아야 한다.
(1) Hemocytometer
• 현미경으로 관찰하며 소량의 세포 현탁액 내 세포의 숫자를 셀 수 있게
도와주는 유리 슬라이드 형태의 기구

As an example: If the total cell count is 145 & DF of the cell suspension is 3
(= original cell suspension 1 + dilution buffer 2), the total suspension volume

(2) Vitamin
• 가장 많이 이용되는 종류는 vitamin B-group (folic acid, biotin, pantothenate 등)
• 일부 배지에는 ascorbic acid (vitamin C)와 a-tocopherol (vitamin E)이 포함
• 일반 배지에 첨가되어지는 혈청은 주요한 비타민 원으로 작용한다. 따라서 혈청의 농도가 낮아지거나
제거되어지면 비타민 첨가량을 증가시키거나 새로운 비타민 종류를 첨가해야 한다.
• 비타민은 대사과정의 보조 인자로 작용하기 때문에 저농도의 비타민은 대사과정을 제한하여 세포 생
존과 증식 속도에 영향을 미친다.
(3) Ions
• 이온은 배지의 삼투압을 결정
• 배지에 사용되는 이온은 Na+, K+, Mg2+, Ca2+, Cl-, SO42-, PO43-, HCO3- 등
• Ca2+와 Mg2+는 부착과 cell aggregation의 보조 인자로 작용하기 때문에 부유 세포 배양에서는 감량
되어 있다.
(4) Carbohydrate
• 대부분의 배지에서는 D-glucose가 L-glutamine과 함께 에너지원으로 작용
• D-glucose는 glycolysis를 통해서 pyruvate로 전환되고 pyruvate는 Krebs 회로를 통해서 에너지를
생산
• 일부 세포주에서는 이 Krebs 회로가 불완전하여 다른 대체 에너지원을 요구하는데, 이때 제일 많이
사용되는 것이 L-glutamine이다.
• D-glucose는 D-galactose, D-fructose, D-mannose와 같은 다른 탄수화물로 대체될 수 있는데, 대
체된 탄수화물의 사용은 배지내 젖산 (lactic acid)의 축적을 방지하여 배지의 산성화를 막음
• sodium bicarbonate / carbon dioxide (NaHCO3 / CO2)는 아래와 같은 반응을 함으로써 세포내
pH를 유지함
• NaHCO3 + H2O → Na+ + HCO3- + H2O → Na+ + H2CO3 + OH- → Na+ + OH- + H2O + CO2
→ increased alkalinity
• 단점 보완
• (2) NaHCO3 를 대신하여 Tris를 사용. 그러나 Tris는 amine을 가지고 있어 화학 반응성이 높고, 세포
증식의 저해제로 작용할 수 있고, 생체막을 통과할 수 있고, 생체막을 통과할 수 있고, 온도에 따라 pH
가 변화되는 단점이 있다.
• (3) pH 6.15 (MES)에서 pH 9.55 (CHES)의 범위를 망라하는 zwitterionic buffer 계열을 도입하였는
데 이에 HEPES가 포함되어 있다.
• HEPES buffer는 37℃에서 pKa 7.31을 가지고 있어 세포 배양 배지에 최적하며 세포막을 통과하지
않는다. HEPES를 배양배지에 포함시킬 경우에는 삼투압을 맞추어주기 위해서 NaCl 농도를 낮추어
준다.
(1) 혈청의 역할
• 혈청(serum)
기본 배양 배지에 주요한 첨가제
광범위한 고분자 단백질, 저분자 영양분, 호르몬, 미네랄, 지방 등을 운반하는 수송 단백질을 제공
단백질 분해효소 (protease)의 활성을 억제하고, pH 조절용 완충제 역할을 하는 등의 장점

• 단백질(protein)
단백질은 혈청의 대부분을 차지하는 성분으로 혈청 내 조성 및 기능들이 완전히 밝혀진 것은 아니
다.
albumin, globulin, 세포 부착 촉진시키는 fibronectin과 fetuin, trypsin을 억제하는
α2-macroglobulin, 그리고 철과 결합하는 transferrin 등이 있다.
(3) 혈청의 성분
• Polypeptide
혈액 채취 직후 원심 분리로 세포를 제거한 혈청보다 자연적으로 응고된 혈청으로 세포를 배양하
면 세포가 더 잘 증식된다.
이것은 혈소판에서 유리되는 polypeptide 때문인데, 이에 속하는 platelet-derived
growth factor (PDGF)이다.
PDGF는 세포 증식을 촉진하는 능력을 가지고 있어 섬유아세포와 신경교의 증식을 촉진한다.
transforming growth factor (TGF)와 같은 혈소판 유리 polypeptide는 상피세포의 증식을 억제
하고 세포분화를 촉진한다.
fibroblast growth factor (FGF),epidermal growth factor (EGF), endothelial growth factor,
insulin-like growth factor (IGF-1, IGF-2) 등이 혈청 내 소량으로 존재
(3) 혈청의 성분
• 호르몬 (hormone)
혈청내에 존재하는 호르몬들은 세포의 증식에 많은 영향을 줄 수 있으며
동일한 호르몬이더라도 세포의 종류에 따라서 서로 다른 효과를 나타낼수 있다.
Insulin은 glucose와 아미노산의 세포내로의 섭취를 촉진하여 세포 증식 촉진 효과를 나타낼 수
있고,
IGF-1 수용체의 활성도에 의해서 그 효과를 나타낼 수도 있다.
태아 혈청에 존재하는 hydrocortosone은 FBS에서 그 양이 일정치 않은데, 세포의 유착과 증식
을 촉진하는 역할을 한다.
그러나, hydrocortisone은 어떤 조건에서는 오히려 세포 증식을 억제하고 세포 분화를 유발하기
도 한다.
(3) 혈청의 성분
• 각종 무기물질(inorganic component)
소량의 미네랄 성분과 철분, 구리, 아연 등은 혈청 단백질에 결합된 상태로 혈청 내에 존재
selenium은 GSH synthetase의 보조인자로서 free radicals를 해독하는데 도움
• 억제 인자(inhibitor)
혈청 내에는 또한 세포 증식을 촉진하는 인자들이 많지만 억제하는 물질들도 존재
세포 증식을 억제하는 인자에는 γ-globulin, TGF-β, 보체(complement) 등이 포함된다.
• 그러나, 무혈청 배지의 주요한 목적은 세포가 생산하는 생리활성물질의 분리정제 과정을 최소화
하기 위한 경우가 많으므로 불필요한 인자들의 첨가는 피하는 것이 좋다.
• 최근에는 철을 수송하는 단백질인 transferrine 대신에 iron citrate, iron chelating reagent를
첨가하여 단백질 성분을 전혀 함유하지 않은 protein-free media도 개발되었다.

- Self renewal: the ability to go through numerous cycles of cell division while
maintaining the undifferentiated state
- Potency : the capacity to differentiate into specialized cell types
- Clonality: ability of a single cell to form many similar cells
- Differentiation: process by which a less specialized cell becomes a more
specialized one

Characteristics
• Exist in small number

Abnormal amplification
• Sigmoidal curve
• Too low baseline setting
• High level of fluorescent noise during early cycles of PCR

• A260/A280 1.8 suggests there is about 70~80% proteins that inhibit PCR

General observation of visible light
• Light source (광원)
• Visible light (from sun) touches an object + reflection + recognize the reflected light with
specific wavelength by eyes
• Ex) imagine all red-colored objects in a dark room
Fluorescence
• Light with a spectrum (Excitation spectrum) touches a fluorephore
• The fluorephore reacts with a specific wavelength (or a specific energy) of the light
• The fluorephore then emits its own light, which corresponds with the wavelength (Emission
spectrum)
