1. Get 100 ml water sampling containing zooplankton in one of the ponds in CPU
campus using a plastic sampling bottle.
2. Fix water sample with 5% formaldehyde or Lugol’s solution (5-10 minutes).
3. Clean the Sedgwick-Rafter counting chamber using tap water and tissue paper.
4. Put fixed 1 mL water sample in the Sedgwick-Rafter counting chamber using a
pipet or medicine dropper. Be sure that the whole area where the grid lines are
found will be covered by the water sample. No bubbles will be formed within the
area.
5. Using the microscope, examine the water sample.
6. Identify the species of zooplankton present.
7. Count and compute the number of population for each species of zooplankton.
Computations:
Number of species = Total counts/mL
8. Make 3 replicates out of the water sample and get the mean.
9. Illustrate the species of zooplankton present in the water sample.
