특히 보관 단계와 그 이후의 배송 단계에 대해 특별한 관심을 두지 않은 것은 잘 알려진
사실이었다. 사실 미국 FDA 규제 환경에서는 보관/배송 등에 대한 제대로 된 가이드가
아직까지도 없는 상황이고 반면에 EU GMP에서는 1994년에 발행된 Good Distribution
Practice(GDP) 가이드[1]가 있었지만 이 가이드는 불과 4쪽짜리 문서로서 충분한 정보를
담고 있지 못했었으며 또한 규제기관 조차도 이에 대한 준수 여부 사찰은 소홀히 여겼던
것이 현실이었다. 1994년판 EU GDP 가이드라인은 2013년 5월에 전면 개정되어
발표되었으며 2013년 9월 8일부로 발효되었다 [2].
영국 MCA를 예로 들면, 2001 – 2002년 사이에 MCA가 실시한 GDP 분야 실사가 총178건
진행되었는데 7개의 중대(Critical deficiency) 지적사항, 161건의 중요(Major deficiency)
지적사항이 확인되었을 정도로 의약품 보관/유통 분야에서 많은 부실이 파악되기에
이르렀다 [3]. 이에 MCA에서는 이 분야에 대한 규제강화를 시작하게 되었으며 이 당시
많은 의약품 도매상들은 제대로 된 규정이나 가이드가 없는 상황에서 규제기관으로부터
그 전에 없던 많은 요구사항들을 업무에 반영하기에는 힘에 부치는 상황이었다. 이에
MCA에서는 Recommendations on the Control and Monitoring of Storage and Transportation
Temperatures of Medicinal Products [4] 라는 일종의 MCA 권고사항을 발표하였는데 Cold
Chain 분야 규제 강화 초반부에 만들어진 중요한 정보들을 담고 있다.

The process of validating/qualifying equipment and storage facilities to prove that they will
create and maintain the temperature and/or humidity they are designed for.

There are not written procedures for production and process controls designed to assure that the drug
products have the identity, strength, quality, and purity they purport or are represented to process.
Specifically, the firm has no written procedure for:
• Drug product storage
• Temperature and humidity warehouse monitoring
Drug products are not stored under appropriate conditions of temperatures so that their identity,
strength, quality, and purity are not affected. Specifically,
• Thermometers at the different (control temperature and humidity) warehouse are not calibrated
and/or periodically verified for accuracy of functionality.
Products were not held at required temperature during shipment. Specifically,
• There are no studies to support the current packaging and shipping practices. Labeled storage
temperature for all manufactured product X is 2 – 8 oC. Packaging and shipping conditions
consist of placing product X in insulated cardboard box(es) wrapped with or without a thin
plastic strip(s) and sometimes with styroform chips (to prevent breakage) and shipped un-
refrigerated to the customer.
Drug products are not stored under appropriate conditions of temperature and humidity so that their
identity, strength, quality, and purity are not affected. Specifically, drug products are stored outside the
recommended storage conditions. For instance:
• Products labeled with warnings to avoid freezing are kept inside a freezer with temperatures
below 5 oC. For example, product X labeled to be kept at room temperature was inside a freezer.
A bottle of product X, labeled “discard if frozen” was also in this freezer. The observed
temperature inside this freezer was below 3 oC.
• The air conditioning system inside the warehouse was off when the inspection started. The
thermometer in the warehouse adjacent to prescription drugs showed a temperature of 27 oC.
The warehouse had several drug such as product X, labeled to be kept between 20 oC to 25 oC.

용적의 약 70 ~ 80 % 정도를 모의 적재한 6 가운데 시험하는 것으로서 실무적으로
어려움이 없었다. 하지만 창고는 그 규모에 있어서 인위적인 적재 조건을 만드는 것이
가능하냐는 것이었다. 아니 가능은 하겠지만 그것이 타당하고 과학적이고 가치창조적인
행위인가에 대한 고민이라고 할 수 있겠다.
Temperature controlled space 예로서 facility, vehicle, shipping containers, refrigerator, freezer
등을 명시하고 있다.

그리고 PQ 과정에서 loaded chamber mapping 이 실시되고 그리고 특별 테스트들 이를 테면
door open test, power failure test 등이 역시 PQ 과정에서 진행된다.

• 소프트웨어 사용 편의성 및 Part 11 준수 – 소프트웨어 자체에 대한 컴퓨터
밸리데이션이 가능하지 여부(굳이 국내에서 미국 20 CFR Part 11 준수
여부까지는 고려하지 않아도 되지만 이는 전적으로 사용자 결정의 몫이다).
일부 메이커 소프트웨어에서는 자동으로 MKT(Mean Kinetic Temperature – 필자가
기고한 GMP Update & Technology Follow-up, Vol. 7 해당 글 참조)를 계산해 주는
기능이 반영되어 있기도 하다.
Step 3과 Step 5에서 확인되고 결정된 장소에 온도/습도 매핑을 실시하기 위하여 데이터
로거를 적절하게 세팅하여 배치한다.

• 데이터 로거 전용 소프트웨어를 사용하여 데이터 로깅 시작시간, 종료시간,
데이터 저장간격 등을 세팅한다. 가능하면 모든 데이터 로거의 시작/종료시간을
동일하게 세팅하여 추후 데이터 처리를 용이하게 하도록 한다.
• 소프트웨어가 21 CFR Part 11을 충족하는 제품이라면 Audit Trail 기능이 반영되어
있으므로 반드시 이 기능을 활성화시켜서 데이터 완결성이 있음을 보증한다.
테스트 기간 중에는 시험자의 특별한 개입이 필요치는 않다. 다만, 중간 중간에 제대로
데이터 측정/저장이 되고 있는지, 이상 데이터 발생은 없는지 등에 대한 중간점검이
필요할 수는 있다. 그러나 중간점검을 위하여 데이터 로거 수거, 데이터 다운로드 등을
해야 한다면 이로 인하여 시험 장소에 빈번히 접근할 경우 시험 결과에 나쁜 영향을 줄
수 있으므로 신중해야 한다. 일부 데이터 로거의 경우 근적외선 무선통신 기능이
탑재되어 있어서 중간점검을 아주 용이하게 해 주는 것들이 있기도 하다.

결론이 도출될 가능성이 상당이 높다. 이것은 다음과 같은 용어로 표현이 가능하다.

4. 매핑에 관한 자주하는 질문들
내부 용적이 2 m3 이하일 때:

그러나 20 m3 초과하는 사이즈에 대해서는 아무런 언급이 없다. 그렇다면 창고의 경우가
이에 해당할 텐데 과연 우리들은 어떻게 해야 한단 말인가 되묻지 않을 수 없다. 필자
역시 이 글에서 이미 설명한 그 이상의 시도를 하지 않을 것이다. 필자 역시 뭔가
확실한 공식이나 룰을 만들어 낼 만큼의 역량을 갖고 있지 못하니까 말이다.

상대습도 센서 개수는 여전히 논란거리 중의 하나이다. ISPE Cold Chain Management
가이드[14]에서는 다음과 같이 설명하고 있다.
“Where relative humidity is a critical factor, then humidity sensors could be located in the same
locations – or fewer locations used with the impact of the temperature considered for the other
locations – as absolute humidity will be very similar throughout the space.”
“If humidity has been defined as a critical quality attribute, and there are no significant source of
humidity in the warehouse, it may be assumed that humidity will equalize within the warehouse. Two
mapping sensors mounted at different heights are suggested, again to be in locations selected to detect
changes from the most significant influences, e.g., door opening.”
IEC 60068 Part 3-6 규격서 [13]에서는 사이즈 구분 없이 습도는 정중아 1곳만 명시하고
있다.

72시간 이상을 하는 것으로 사내 기준을 설정한 회사도 있다.
그런데 아일랜드 의약품 규제기관인 HPRA가 발행한 Guide to Control and Monitoring of
Storage and Transportation Temperature Conditions for Medicinal Products and Active Substances
[16]에서 다음과 같은 내용을 발견할 수 있다.
“Warehouses should be temperature mapped in the empty and full states to determine the temperature
distribution under extreme of external temperature.”

초기(initial test after warehouse construction) 시점에서 empty/full 상태 모두를 시험하라는
의미는 아니라고 해석할 수도 있다.
ISPE에서 발행한 Cold Chain Management 가이드 [14]에서 이에 대한 명확한 언급이 있다.
“It is normal practice to use the air temperature as the reference source for the stored
temperature/hence the worst case representation of the stored product temperature.”
그러나 door open test, power failure test와 같은 특별한 시험에서는 공기 온도를 측정할 경우
현실적으로 신뢰할 만한 데이터를 얻을 수 없다. 그 이유는 이를 테면 1000 리터 용적을
갖는 안정성챔버를 가정할 때 문이 열린 후 얼마 지나지 않아서 챔버 내부의 공기
온도는 외기의 영향으로 바로 상승(또는 하강)하게 된다. 상대습도를 측정해야 한다면
상대습도는 온도보다 더 빠르게 변화한다. 따라서 이들 시험에서 공기 온도를 측정할
경우 의미 있는 데이터 확보가 안되므로 따라서 제품 온도를 기준으로 시험해야 한다.

논의되기도 한다. 이를 테면 혈액보관냉장고의 경우 우리나라 KS 산업규격에서 8 온도
기록용 센서는 문을 열 때 외부 공기에 영향을 직접 받지 않도록 글리세린이 담겨 있는
병 내에 집어 넣도록 되어 있다. 이 부분에 대해서도 ISPE가 발행한 Cold Chain
Management 가이드 [14]에서 제시하는 방법은 2가지이다.

추가 글:
Recently, I've heard that a company have performed temperature mapping studies for cleanrooms. In
general refrigerators, stability chambers, incubators, etc (i.e., environmentally controlled chambers)
are undergone the temperature mapping studies under empty and full loaded states in order to
determine temperature distribution profile within the chamber. Also warehouses should be temperature
mapped.
But I've never seen any technical articles or GMP regulations/guidelines that temperature mapping
studies should be required for each cleanroom. The story I heard is that a company placed at least 10
data loggers at each cleanroom and temperature data was collected for at least 24 hours. For doing this,
the company used lots of resources, time, and money. I’m not sure the company specified that the
temperature mapping should be performed in their URS. Whatever the truth, I think this practice is
very weird and wasteful non-value added activity.
As you well know, ISO 14644-3 mentions two different temperature tests. The first is a general test
and the second is a comprehensive test. Those test procedures are derived from IEST-RP-CC006.3
(Testing Cleanrooms). Are those tests enough to demonstrate temperature control within a cleanroom?
Is cleanroom temperature mapping just like as environmentally controlled chambers considered as a
sort of industry general practice? Do you know any FDA 483 citation on this matter?
The reason environmentally controlled chambers and warehouses are temperature monitored is to
determine where temperature sensitive material can be store. During manufacturing of product
normally temperature is controlled by other means than room environment. If room temperature is the
only way of controlling product temperature. I would monitor product temperature to control the
HVAC temperature and not control HVAC temperature and hope I maintain product temperature.

I have not seen any 483s for not temperature mapping a clean room.
Temperature mapping is for areas where product can sit (i.e. be stored) for extended periods.... more
than just a few hours.
Most pharma cleanroom temperatures are maintained well within the limits established for the product.
A few aseptically processed products (such as insulin) may be processed (filled) cold, but product
stability data have should already have investigated the effects of short residence in a room maintained
at "worker comfort" levels and established a maximum residence time.
Short answer.... temperature mapping should not be an expectation for a non-storage area.
First port of call should be the HVAC URS. I've only ever requested a comprehensive test in larger
cleanrooms or where temperature was considered product quality critical. Is temperature at the facility
controlled for occupant comfort or product quality?
As previously mentioned, temperature mapping is normally reserved for product storage such as
freezers, cold rooms, stability chambers etc...
Baring processing under special conditions (e.g. in cold rooms) due to temperature sensitive products;
I have not seen any 483's or EU observations on this (not to say it hasn't happened) nor have I ever
issued an acceptable temp variability spec in a room that didn't have special product conditions.
