4.Nitrous Acid Test


Take small quantities of primary,
secondary and tertiary amines
and put in a test tube a, b and c
respectively. Using a spatula,
take a small amount of sodium
nitrite in a test tube and add
distilled water. Shake the test
tube well and cool the solution in
ice bath. Add small quantity of
HCl to test tube a, b and c and
shake the test tube well. Cool the
Primary amine Nitrous Acid test tubes in ice cold water. Add
Secondary amine the sodium nitrite solution to test
Tertiary amine tube a, b and c.

Results:
Primary amine produces bubbles
(nitrogen gas) (RNH2 + HONO
 ROH + H2O + N2)
Secondary amine formed yellow
oily layer (R2NH + HONO 
R2N-NO + H2O).
Tertiary amine formed soluble
nitride salt (R3N + HONO 
R3NH+ ONO-).
5. Hinsberg Test Take small quantities of primary,
Primary amine secondary and tertiary amines
Secondary amine and put in a test tube a, b and c
Tertiary amine respectively. Using a dropper,
add a small quantity of 25% HCl
to test tube a ,b and c. add small
quantity of distilled water to each
test tube. Add Benzene sulphonyl
chloride to each test tube and
shake well. Cool the test to in ice
water for few mins.

Results: Secondary and tertiary


amines turned translucent while
the tertiary remains crystal clear

Tertiary amine formed N-Alkyl
benzene sulphonamide
(insoluble).
Secondary amine- No reaction.
Tertiary amine reacts with HCl
and formed soluble ammonium
salt.
B. 1. Biuret Test Take a small amount of egg
Egg Albumin dispersion albumin dispersion and gelatin
Gelatin dispersion dispersion put in a separate test
tubes a and b. add small amount
of NaOH to the 2 test tubes a
and b. add few drops of 1%
NaOH and CuSO4 CuSO4 to the 2 test tubes and
heat the two test tube for few
mins over the burner.

Result: the solution turned violet


due to the formation of Biuret.
2. Ninhydrin Test Take a small amount of egg
Egg Albumin dispersion albumin dispersion and gelatin
Gelatin dispersion dispersion put in a separate test
tubes a and b. add a few drops
of Ninhydrin solution to the test
Ninhydrin Solution tube a and b. boil the test tubes
over the burner.

Result: 2 test tubes turned dark


blue.
C. 1. Dragendorff’s Test Mix 0.85 g Bismuth oxynitrate,
Tonic Water 10 ml acetic acid (99-100%) and
Extract from poppy seed 40 ml Dis. water in a beaker.
Agitate to dissolve. In a separate
beaker, mix 0.4 Potassium
idodide per ml of dis. water, 2 g
Potassium iodide and 5 ml dis.
water. mix a 1:1 of the two
Dragendorff’s reagent solutions. The solution turned
dark yellow (Dragendorff’s
reagent).
Put 2 ml of tonic water in a test
tube and shake it to remove the
CO2. Add 1 ml of Dragendorff’s
reagent in a test tube containing
tonic water.
Result: yellow solution becomes
cloudy due to the formation of
precipitate.
Put small amount of poppy seeds
and water in a mortar then add 2
drops of cons. HCl. crush the
seeds with the pestle. The
mixture become a suspension
with a white solid. Filter the
mixture.
Result: yellow solution becomes
cloudy due to the formation of
precipitate
2. Meyer’s Test Sample containing alkaloid +
Alkaloid sample Mercuric Chloride, Potassium Mayer’s reagent  creamy white
Iodide dissolved in distilled precipitate.
