京都チーム成功 マウスの子も誕生
様々な組織や臓器に変化できるマウスの iPS 細胞(新型万能細胞)から卵子を作ることに、
斎藤通紀(みちのり)・京都大学教授らの研究チームが世界で初めて成功した。この卵子か
ら健康な子が誕生した。
特殊な細胞培養法を用いているため、直ちに人に応用することはできないが、チームは昨年、
マウスの精子作成にも成功しており、将来的には不妊症研究に利用できる可能性がある。 5
日の米科学誌サイエンス(電子版)に発表する。
斎藤教授らはまず雌のマウスの皮膚から iPS 細胞を作製、卵子や精子のもとになる「始原生
殖細胞」を作った。この細胞だけでは卵子ができなかったため、別の雌のマウスの胎児から、
卵子を育てる卵巣のもとになる体細胞を取り出し、 iPS 細胞から作った始原生殖細胞と一緒
に培養した。
2種類の細胞がくついた塊を成熟した雌の卵巣に移植すると、4 週間後に始原生殖細胞が卵
子に成長した。卵巣のもとになる体細胞が始原生殖細胞を卵子に成長させる物質を出してい
るらしい。さらに、できた卵子に健康な雄から採取した精子を体外受精させ、約 10 匹の成熟
した雌の卵管に移植したところ、2 匹から健康な子供のマウス 3 匹が誕生。その子供の世代
も生まれた。

Baby Mouse Born


A research team of Kyoto University staff headed by Professor Michinori Saito have for the first time,
been successful in growing an egg cell from an iPS cell, a new type of pluripotent cell which can
develop into many different types of organ and tissue. A healthy baby mouse was born from this egg
cell.
This cannot be applied to humans as yet, as special culture method is used, but as the team were last
year also successful in constructing sperm cells there is a possibility it could be used in fertility
research in the future. The discovery is announced today in the electronic edition of the US academic
magazine Science.
Saito and his colleagues first made an iPS cell from the skin of a female mouse, from which they
formed a “basic reproductive cell” which can develop into a sperm or an egg cell. An egg could not be
formed from this cell alone, so from another mouse embryo, they took somatic cells which later
develop into the ovaries which nurture the eggs and cultured them together with the basic reproductive
cells which had been made from the iPS cells.
The clump of both types of cells was transferred into the ovary of a mature female, and after 4 weeks
the basic reproductive cells had developed into eggs. It seems that the ovary-forming somatic cells emit
a substance which develops the basic reproductive cells into eggs. Furthermore, when eggs made this
way were fertilised in vitrio with sperm from healthy males and transplanted into the fallopian tubes of
ten mature females, two of those females gave birth to three healthy mice. These young also gave birth
to their next generation.
