Experiment No. 2

Plant Tissue and the Multicellular Plant Body


Objectives:
At the end of this experiment, students are expected to:
1. Describe the different plant tissues and its function;
2. Demonstrate characterization of the plant tissues of monocot and dicot by
differential observation under microscope;
3. Create a microscope slide samples of plant tissue.
Materials and Reagents:
Microscope Permanent slide of Clear Nail Polish
cross-sectional corn
(Zea mays) stalk/stem.

Fresh leaf Glass slide and cover Transparent Tape


slip

Permanent slides of Scalpel or razor blade Distilled water


parenchyma, collenchyma,
& sclerenchyma

Procedure
A. The Ground Tissue System
1. At low microscope magnification, observe parenchyma, collenchyma
and sclerenchyma permanent slides.
2. Draw and label.

B. The Vascular Tissue system


1. At low microscope magnification observe the slide of the collateral
fibrovascular bundles in the corn stalk cross section.
2. The fibrovascular bundles are arranged randomly. Find a
fibrovascular bundle and observe it carefully.
3. Draw and Label
The bundle is surrounded by uniform cells with thick walls, painted in
red, sclerenchyma. Closer to the center of the stem there are three spiral and
annular vessels surrounded by woody parenchyma (xylem). On the outside of
the larger vessels there is phloem represented by the sieve tubes and
companion cells. There is no vast parenchyma in the phloem, which is typical
for monocotyledonous plants, including corn. Notice that the xylem in the
section surrounds the phloem by a semicircle, which is also characteristic for
the monocotyledonous plants. Xylem is located close to the center and the

phloem – to the periphery. Another feature of the monocotyledonous plants is


the absence of the secondary meristem, cambium.
C. The Dermal tissue system
1. On the bottom of the leaf, paint a 1-inch square section of clear nail
polish. Allow to dry.
2. Place clear tape over each section of nail polish and carefully remove
from the leaf. This will give you an impression of the epidermis of the
leaf. Fix the tape to a microscope slide.
3. With the microscope on the lowest setting, focus your slide to see the
stomata. Once you have a good view, switch to the next magnification.
Look at the stomata from each magnification available. Determine at
which magnification the stomata are the most easily viewed.
4. Draw and label.

- Xylem transports water from the root throughout the different plant part
and also providing structural support while phloem transports organic
compounds from photosynthesis to the different plant parts.

- The role of meristems in plants is to create new cells, tissues, and
organs so that the plant itself continues to grow and survive.

- Plants continue to grow as long as they are alive, and there is no limit to
their growth. For animals, growth eventually ceases when it reaches a
certain point.
