OBJECTIVE

To study
the polen germination on stigma through apermanent slide.
MATERIALS REQUIRED:
Fresh flowers, microscope, slide, cover slips, sucrose, boric acid, potassium nitrate, magnesium sulphate,
beakers, etc.
PROCEDURE
1. Prepare anutrient medium by mixing 10 gm sucrose, 10 mg boric acid, 30 mg magnesium sulphate and 20
mg potassium nitrate in 100 ml of water.
2. Take a few drops ofthis nutrient medium on a slide and dusta few pollen grains from the stamen on it. Leave
them undisturbed for about 10 minutes.
3. Observe the slide under a microscope.
OBSERVATIONSAND CONCLUSION
The pollen grains germinate in a nutrient medium. The tube cell enlarges to come out of thepollen grain through
one of the germ pores and forms a pollen tube. The tube nucleus reaches the tip ofthis pollen tube. The generative cell
also passes into it and divides into two male gametes.
