THINGS TO REMEMBER

• Percentage solutions: % means = amount (usually in grams) per 100ml


For example 20% SDS power = 20g of SDS power in 100ml
- w/v units = g
- v/v units = volumetric units
- w/w units = g

• Dilution(D) is always < 1 & Dilution factor(DF) > 1

Example: A 10 fold serial dilution was performed:

The total dilution volume is 1ml and you plated 100ul; Dilution = 100/1000 = 10-1

If you plated 2ul then your Dilution would be = 2/1000 = 1/500

D = v(stock)/v(working solution)

D = c[working solution]/c[stock]

DF = v(working stock)/v(stock)

DF = c[stock]/c[working solution]

• Molarity
- C = n/v or c=m/Mw*V
- C units = M or mol/L N = m/Mw

WORDING:

1) Weight out(if in g); measure out (if in volumetric units)


2) If solute if a solid then do not fill till final volume; first dissolve solid solute in ~60% of the
final volume of solvent.
3) Adjust pH (if pH needs to be adjusted)
4) Fill up to final volume of…

NB!!!! When you prepare a diluted acid, NEVER add water to the acid! Add acid to water
INSTEAD.
