Procedure for the ACQUITY UPLC

Verify completion of the operational qualification procedure for the ACQUITY UPLC® sample
organizer (SO) by completing Form 8. Initial the Not Installed line if the SO option is not
installed on your ACQUITY UPLC system. If you require information about how to perform any
of the steps in this form, see page 2.

Testing the Sample Compartment Temperature

Recommended Materials
• Calibrated thermometer probe, with readout device
• Tape, to hold the thermometer probe in place
To test the sample organizer’s sample compartment temperature:

Note: The intent of step 1 is to position the thermometer probe wire in the sample organizer.
Depending on the design of the thermometer probe you use, alternate methods can be
used.

1. Open the sample organizer’s door and position the thermometer probe wire on the
X-transfer shuttle mechanism so that the probe tip extends one-inch (2.54 cm) beyond the
rear of the mechanism. Use tape to hold the probe in place. (See Figure 1-1.)

Note: Avoid placing the thermometer probe wire in the center of the X-transfer shuttle
mechanism, to prevent activating the plate over-travel sensor.

2. Close the sample compartment door.


3. In the ACQUITY Console screen for the sample organizer heater/cooler, specify a set point
temperature.
2. Select your standard operating temperature or a temperature as low as 4 °C (not to exceed
20 °C below ambient), and record this set-point value in Form 8.
4. Allow sufficient time for the plate compartment temperature to reach and stabilize at the set
temperature.
3. In Form 8, record the temperature you observe from the thermometer probe.

Note: The time required to reach the set temperature varies depending on the difference
between the set and ambient temperatures. For example, the plate compartment can
require up to 90 minutes to reach 4 °C when starting from room temperature.

5. When the test is completed, remove the thermometer probe wire from the sample
organizer.

Testing the ACQUITY UPLC Sample Organizer Plate


Positioning

Recommended Materials
Four SBS/ANSI sample plates
To test plate positioning of the sample organizer:
1. Ensure there are no plates in the sample manager.
2. Load the sample organizer with four ANSI sample plates.
Requirement: Load one plate at the top position, another at the bottom position, and the
remaining two plates at random positions between the top and bottom.
3. Record in Form 8 the four plate positions used.
4. Close the sample compartment door.
5. On the ACQUITY Console’s sample organizer screen, select Configure > Scan and
Store Shelf Layout.
6. On the right-hand side of the ACQUITY Console screen, select Sample Organizer >
Interactive Display, and then click the Control icon, to activate the sample organizer
controls.
7. On the ACQUITY Console screen, click the label that designates the position of the
bottom shelf that was loaded in step 2.
8. In the Transfer to Sample Manager dialog box, click Yes, and ensure the plate on the
designated shelf is transferred from the sample organizer to the sample manager.
9. On the ACQUITY Console screen, click the label that designates the position of the
bottom shelf.
10. In the Transfer to Shelf dialog box, click Yes, and ensure the plate is transferred from the
sample manager to its original shelf position in the sample organizer.
11. Repeat step 7 through step 10 for the other three sample plate positions that were
loaded in step 2.
12. Record the results in Form 8. To pass this test, each of the four plates must correctly
transfer from the sample organizer to the sample manager, and then back to their
original position in the sample organizer.
