Outline Possible causes and countermeasures for the HA-8160V abnormal baseline in chromatograph is explained in | this document. Optical unit cell washing or optical unit assembly related parts replacement may be effective in | some cases, 2. Details In HA-B180V, when measurement value is supposed as abnormal, baseline drift because of dirt in flow channel is considered a problem cause. Baseline in chromatograph can be checked by the instrument diagnosis function. Enter the Service Mode and select the menu below, then execute Analysis section check. Baseline is printed out automatically. Next, check the printed result comparing with the normal waveform and the following five check points. Check baseline using Analysis section check measurement. (Service mode>>8.Instrument diagnosis menu>>4.Analysis section check) If the value exceeds the acceptable range below, Drift baseline, Noise detected, or T490:Background occurs. Attention is needed also when waveform is around the threshold. Note that the baseline vertical reference position is one division higher from the horizontal axis. Normal waveform Points to check on baseline 1) Baseline shift while Eluent-CV is fed to the column. (Only for Variant Mode) Check that the baseline shift while Eluent CV solution is fed does not exceed +/-0.5 division shown in the examples below. (In case the waveform looks like the examples 1, 2, 3, 5.) — Optical cell characteristics degradation induced by protein stain adhering to the optical unit after instrument long use or cell dry-up due to solution leakage. Cause 2) Light beam misalignment. (In case the waveform looks like the examples 1, 2, 3, 5.) + Optical cell deformation caused by flow channel higher pressure induced by optical cell output resistant pipe clogging by mechanical shock or long term use, or looseness of fixing screws for interference filter block or optical unit cell. Cause 3) Optical transmission wavelength shift (In case the waveform looks like the examples 1,2,3,5,) — Lens or interference filter characteristic degradation due to dust or soot adhering to lens or dew condensation on lens, Cause 4) Air bubble mixing (In case the waveform looks like the examples 2, 4, 5.) — Air mixing or fluid leakage from tube joint portion or optical cell Cause 5) Dirt of Eluent (In case the waveform looks like the examples 1, 2,3.) — Incase Eluent is dirty, ete. [Countermeasures] ‘When baseline drift occurs, following countermeasures can be taken. Countermeasure 1) Check for air bubble in the drain tube. (Cause 4 is suspicious.) — Check for air bubble in the instrument OUT tubing (one connected to ‘D1 in the rear) during measurement. if air bubbles enter the tube, joint looseness or fluid leakage from optical unit assembly or degasser maifunction can be considered. When air bubble mixing or fluid leakage is found, check the tube connection and fluid leakage from a cell. Countermeasure 2) Replace Eluent ( Cause 5 is suspicious.) — Eluent may get dirt. Replace Eluent with a new one. Also, replace fluid inside Eluent chamber. (Cause 1 is suspicious.) —Wash the optical unit cell with Cell Washing Kit (Product code:80386) (Reference: Service manual Chapter? 1.1) Washing the optical unit cell) Countermeasure 4) Replace the optical unit, (Cause 1,2,3 is suspicious) —ln case the cause 3 hits, disassemble and clean the interference filter and the cell. If disassembling and cleaning is difficult, optical unit assembly or interference filter plus cell Note that countermeasures other than above may be required depending on the situation. <> When the measurement value shows abnormality, column replacement or low pressure tube line washing may be effective.
