Practical: To perform qualitative test for analysis of

phenolic compounds

1. Ferric chloride test for phenols
Chemical names: ferric chloride, water, plant sample

Procedure
• Take 2 ml plant sample in the test tube.
• Add 2 ml of ferric chloride solution (0.1 %) was into it.
• A deep bluish green solution indicated presence of phenols

2. Lead acetate test for tannins

• Take 1 ml aliquot of sample into the test tubes,


• After this add 5 ml of 1% lead acetate solution into it.
• The formation of white precipitate indicated the presence of tannins

3. Ammonia test for flavonoids :
Chemical names: ammonia, conc. sulphric acid, plant sample.
Procedure
• Take 2 ml of plant sample then add 3 mL of dilute ammonia into it.
• To this add of 1 mL concentrated sulphuric acid (H2SO4).
• Yellow coloration in test tubes showed the presence of flavonoids

Sodium hydroxide test for flavonoids :
Chemical names: sodium hydroxide, test tube, plant sample.
Procedure
• Take 2 ml of plant sample into the test tube.
• Then add 3 mL of sodium hydroxide into it.
• Yellow coloration in test tube showed the presence of flavonoids.
