Worklog Paper

Created by
Nichapha Chayarangkan Grade 11 Section 1101

Presented to
Mr. Monchai Kijbumrung
Remarks: * I starts intern before time because of my mentor research need to be done as fast as it can be *

Dates Activities Number of Hours

22 September 2019 - Meet mentor and discuss our 2 hours

responsibilities.

24 September 2019 - Get the scientific research paper 3 hours

about rice root
- Go to the lab for help and learn how
to use the equipment in the lab

25 September 2019 - Read and understand the research 4 hours

paper and summarize to mentor
- Cut paper for use in the experiment
and then sterilized it

26 September 2019 - Discuss paper with a mentor 2 hours

- Know the plan and hypothesis of this
research project.
- Mentor explains the method of the
experiment

30 September 2019 - Explain the general skills that use in 2 hours

lab and experiment
- Do the experimental design
- Do the randomized of every
type of rice

1 October 2019 - Do the germinating seeds and roll 4 hours

system
- Classified each type of rice
- Mix distilled water with Haiter
together and pour into the rice for
removing the bad rice
- Bring the sterilized paper to the tray
that fills with CaSO4
- Put each of the rice on the paper and
slowly roll the paper until the paper
end then continuously do it until 16
seeds. Repeat this step in every type
of rice
- Put in the cup and cover with plastic
bags
- Put it into the darkest area for 1-2
weeks

2 October 2019 - We went to the camera center at 6 hours

Mahidol in Phayathai to take a
picture of the nut and corn roots
used in the experiment.
- Learn how to use the camera
- Adjust lens
- Move the glass plate to see
the best part for taking the
picture
- Dyed root sample and take a picture
- Eliminate fungus in the roots (In
case, it sticks to the root tightly)
- Take a picture of the root by using
the microscope and specific program
to analyze the root

3 October 2019 - Practice equipment in the lab 2 hours

- Help other senior in the lab to make
more used to with the equipment

8 October 2019 - Calculation the portion of the 3 hours

substance for using in the hydro
water
- Get the rice out and keep it in the
specific place for preventing the
animal and surplus light

28 October 2019 - Insert numbers of corn and nuts 2 hours

roots in meteor excel for using in the
future

29 October 2019 - Do the Hoagland water as a nutrient 4 hours

for rice
- Prepare a stock solution for each
substance, for example, CaNO3 1 M
- Pipette stock solution of each
substance according to the volume
formula and mix in the volumetric
flask
- Fill the distilled water in volume
metric flask until full
- Mix 10 of the substances into a
beaker then pour into a conical flask
- Stir the substance in the hot plate
stirrer that used a magnet to stir in
the substances
- If Hoagland is too intense, we will
add more distilled water to it.
- We use ¼ and ½
concentration
- Adjust pH value of solution until it
reaches 6.5by using KOH which is
base and HCL which is acid
- Divide the 3 beakers at the same
amount
- Before using the Hoagland, we need
to bring it to the autoclave for killing
the germs

31 October 2019 - Place the bottles in the greenhouse 1 hour

4 November 2019 - Changing water of every bottle of 2 hour 30 minutes

rice

6 November 2019 - Changing water of every bottle of 2 hour 45 minutes

rice
- Investigate the changes and record
the changes

8 November 2019 - Changing water of every bottle of 2 hours 30 minutes

rice

11 November 2019 - Help mentor to create and write the 2 hours

envelope to pack the rice in

12 November 2019 - Last time changing water of every 3 hours

bottle of rice
- Investigate the changes and record
the changes

14 November 2019 - Collect the bottle of rice from the 7 hours

greenhouse back to the lab
- Separate each type of rice and
classify into the row of Nitrogen (N),
Phosphorus (P), Carbon (C),
Potassium (K)
- Rearrange the row of the good bottle
to the worst bottle in each nutrient
- Cut the roots
- Measure the height and count the
leaves numbers of each rice
- Softly fold it and put into scientific
digital scales
- Record the number of fresh weight
and height then put it into the
envelope
- Pour the alcohol in the bowl and put
the root in the bowl and find the main
root then cut it
- Put the main root that already cut
into the bottle of alcohol to preserve
in a long period

19 November 2019 - Weigh the dry weight of rice and nut 4 hours
- Record on the envelope and give it
to a mentor to record more further

3 December 2019 - Mentor teaches how to use ImageJ 3 hours

program for counting the number of
root hair and measure the length of
the main root

4 December 2019 - Do the image analysis in C1 sample 2 hours
