effect of teamwork. It involves the co-ordination of many people
around me. The project was a challenging task for me.

changed the complexity of our project into a perfect and smooth

INTRODUCTION
1. Antibiotic - An antibiotic is an agent that
either kills or inhibits the growth of a
microorganism. The term antibiotic was first
used in 1942 by Selman Waksman and his
collaborators in journal articles to describe any
substance produced by a microorganism that is
antagonistic to the growth of other
microorganisms in high dilution. This definition
excluded substances that kill bacteria but that
are not produced by microorganisms (such as
gastric juices and hydrogen peroxide). It also
excluded synthetic antibacterial compounds
such as the sulfonamides. Many antibacterial
compounds are relatively small molecules with
a molecular weight of less than 2000 atomic
mass units.
2. Importance of study of antibiotics - Antibiotic
resistance is becoming more and more
common. Antibiotics and antimicrobial agents
are drugs or chemicals that are used to kill or
hinder the growth of bacteria, viruses, and
other microbes. Due to the prevalent use of
antibiotics, resistant strains of bacteria are
becoming much more difficult to treat. These
represent a threat to public health since they
are resistant to most commonly used
antibiotics. Bacteria are adapting to these
antibiotics making them ineffective means for
treating these types of infection. For example,
Staphylococcus aureus have developed a single
DNA mutation that alters the organism's cell
wall. This gives them the ability to withstand
antibiotic cell disruption processes
Aim – To Study the Effects of
Antibiotics on Micro Organisms

Requirements
Potato ,agar, dextrose , distilled water ,
four different types of antibiotics (such
as penicillin , streptomycin ,
aureomycin , tetramycin ) , syringe ,
oven sterilized petri dish , flasks ,
beakers , pipettes , garden soil , glass
marker pen , etc.
PROCEDURE

A. Preparation of culture Medium -


1. Potato Dextrose Agar (PDA) Medium. Take
200 g of peeled potato chips. Boil them with
500 ml of water in a beaker for 15 minutes.
Squeeze the potato pulp thus obtained through
a muslin cloth and keep it in a flask. Take 20 g
of agar in a beaker and warm it with 500 ml of
water .Mix both the solution of potato and agar
and add 20 g dextrose to it. Thus one liter of
PDA medium is prepared.
Autoclave the medium at 15 pounds pressure
for 15 minutes.
B. Effects of antibiotics on microorganisms in
soil -
i) Take 2 g of soil and dissolve it in 10 ml of
water in a beaker. Let the soil particle settle
down.
ii) Take 5 oven sterilized petri dishes and pour 1
ml of soil suspension in each of the plates. Now
pour 1ml of the four antibiotics separately in
four petri dishes with the help of syringe, and
mark them with marker pen. Leave the fifth
petri dish without antibiotic to serve as control.
iii) Pour PDA in each of the petri dish and mix
the suspension by rotating the petri dish. Leave
the petri dishes undisturbed at a warm place
for an hour.
Iv) Observe the step by step changes and make
note of it .
Observation
Colonies of micro- organisms appear on the
culture medium, count the colonies in each
petri dishes and present your observation in
the following table:

The effect of different antibiotics on the
microorganisms can be assessed by counting
the number and size of the colonies growing in
the petri dishes.

Conclusion
Penicillin and Tetramycin were most effective
antibiotics against microorganisms in soil.
