The document is a biology investigatory project on DNA fingerprinting by Ishita Biswas, a class 12 student. It summarizes the key steps in DNA fingerprinting, which was developed in 1984 by Alec Jeffreys. DNA fingerprinting is a technique used to determine a person's identity based on unique nucleotide sequences in their DNA. It involves isolating DNA from a sample, digesting it with restriction enzymes, separating the fragments by size via electrophoresis, and analyzing the fragments to obtain a unique fingerprint.

CLASS-12 BOARD ROLL NO.- SESSION-2022-2023 Self Certificate This is to certify that I, Ishita Biswas of class XII F of ARMY PUBLIC SCHOOL, BARRACKPORE have solely and exclusively done this investigatory project pursuing essential vital assistance from my teacher and others having knowledge concerning the subject matter. I have also taken references from the web in due course to formulate a successful project. The basic of the project has been put together on bonfire grounds and the allied information is truly genuine. Certificate This is here to inform that Ishita Biswas student of class XII F has successfully completed the project titled "DNA Finger Printing" under the guidance of Mr. Mainak Sengupta (subject teacher biology) during the year 2022-23 in fulfillment of the biology practical examination conducted by AISSCE, New Delhi. Acknowledgement In the accomplishment of this project successfully, many people have owned upon me, their blessings and the heart pledged support, this time I am utilizing to thank all these people, those who have been concerned with this project.
I would like to thank our principal Mrs. Moitreyee
Mukherjee and biology teacher Mr. Mainak Sengupta whose valuable guidance has been one that helped me patch this project and make it full proof success, his suggestions has served as the major contributor towards the completion of this project.
Lastly, I would like to place my sincere to all who
helped me directly or indirectly to complete this project on the time to my family whose appreciation and motivation persuaded to give my best. DNA Fingerprinting DNA fingerprinting is a laboratory technique used to determine a probable identity of a person based on the nucleotide sequences of certain regions of human DNA that are unique to individuals.
The technique was developed in 1984 by British
geneticist Alec Jeffreys, after he noticed that certain sequences of highly variable DNA (known as minisatellites), which do not contribute to the functions of genes, are repeated within genes. Jeffreys recognized that each individual has a unique pattern of minisatellites (the only exceptions being multiple individuals from a single zygote, such as identical twins). The procedure for creating a DNA fingerprint consists of first obtaining a sample of cells, such as skin, hair, or blood cells, which contain DNA.
In Jeffreys’s original approach, which was based
on restriction fragment length polymorphism (RFLP) technology, the DNA was then cut at specific points along the strand with proteins known as restriction enzymes.
DNA fingerprinting steps:
Isolating the DNA. ↓ Digesting the DNA with the help of restriction endonuclease enzymes. ↓ Separating the digested fragments as per the fragment size by the process of electrophoresis. ↓ Blotting the separated fragments onto synthetic membranes like nylon. ↓ Hybridizing the fragments using labeled VNTR probes. ↓ Analyzing the hybrid fragments using autoradiography.
