Dạ em chào anh Thanh ạ,

Em là Phúc, hồi đầu năm nay em có được anh Sơn giới thiệu gặp anh để trao đổi về cơ hội làm
việc tại ĐH Công nghiệp ạ. Như em đã trình bày về tình trạng công việc của em trong lần nói
chuyện trước với anh, em còn cần qua Bỉ một đợt khoảng 6 tháng nữa để làm nốt các phân tích
cần thiết và bảo vệ luận văn Tiến sĩ. Khoảng vài tháng nay em đã cố gắng tìm nguồn kinh phí
học bổng để sang Bỉ hoàn thành nốt chương trình học Tiến sĩ của em tuy nhiên đến nay vẫn
không thành công.
Hiện tại em đang đứng trước quyết định bỏ tiền túi để trang trải chi phí qua Bỉ. Số tiền cần bỏ ra
khá lớn nên em đang khá đắn đo. Chính vì vậy mà em rất muốn tìm hiểu kỹ hơn về cơ hội kinh tế
của công việc giảng dạy tại ĐH Công nghiệp. Rất mong anh giúp em giải đáp một số thắc mắc
sau:

Dear Bart, Maarten and Dinh,

As you already know, the funding for my PhD project ended as of the end of last year. Therefore,
I will stay in Vietnam for the time being and apply for the scholarship from Vietnam National
Foundation for Science and Technology Development (Nafosted). For that reason, the planning
of my PhD will be changed. I hereby send a plan of the rest of my PhD. Could you kindly take a
look and give some feedback if necessary.

Time Task
1/1/2019 – 15/1/2019 - Writing and submitting CA paper (important for CV applying
for fellowship) *

- Analyse CI samples for Vitamin C and malondialdehyde
content.
- Write and submit paper on the protocol for aroma analysis of
dragon fruit
- Develop method to analyse dragon fruit image (using ImageJ)
- Write the Introduction/Literature review/Materials and Methods
for my PhD thesis

June/2019 – (In Belgium)
December/2019 - Analyse samples from the CI experiment (aroma and fatty acid

Below are some further remarks I made for the table:


* Regarding manuscript of CA paper:
Since the waiting time for the fellowship result normally takes at least 3 months, it is important
that we submit the CA paper asap to add it into my CV for the application file. From 23/1/2019
to 29/1/2019 is the Lunar New Year holiday in Vietnam, it would be ideal to submit the
application documents before 23/1/2019. Therefore, Maarten, could you kindly prioritize the
editing of the CA manuscript?

** Regarding shipping of samples to Belgium:


Senne needs the aroma samples from his respiration experiment to be sent to Belgium. Dinh & I
think we should ship some samples of my CI experiment together so I can analyse for aroma and
fatty acid later in Belgium and so that we make good use of this delivery. Although the total
number of samples is huge, after some discussion, we selected interesting samples which
developed translucency to be sent. The number of 15 mL falcon tubes that we plan to send is
648. Together with 72 tubes of Senne and a few samples that Thao also want to sent (about 15
tubes), we have a total of roughly 750 tubes. Still, Maarten, could you kindly plan some time so
that you and I can discuss about the efficiency of this shipment, via Skype, maybe within this
week?

I also want to suggest something new. For the respiration result of Senne’s experiment, I think
they could be combined with the aroma analysis result (which he will perform) so that I can
write another paper on the effect of local ambient temperature on respiration of dragon fruit.
Since I have the experience of writing the first paper on respiration, I think this second will cost
me much less time. Maarten, do you think this is doable?

The student is doing a PhD at two universities at a time and he only gets residence permits for 6
months at a time. Indeed he/she was issued a residence permit but that is not valid anymore.

Dear Maarten,

First to rationalize the sampling of dragon fruit, I’ll present with details how we end up with the number
of approximately 4000 tubes.

First we have fruit harvested at 7 different orchards. This is to study the effect of fertilizer, temperature,
rainfall, tree age, soil type on chilling injury. For each orchard, the fruit will be stored at 2oC or 10oC for
25 days, then transferred to 18oC and stored for another 8 days (in total 33 days). During storage, fruit
are sampled at day 8, 16, 25, 27, 29, 31 and 33. We also sample at day 0. For each sampling point, we
sample 4 fruit, serving as 4 biological replicates. Since the chilling injury symptoms may occur both on
the peel and in the pulp, for each fruit we sample separately the pulp tissue (for further analysis on
reducing sugar, vitamin C, MDA, fatty acid), the peel tissue (for further analysis on reducing sugar,
vitamin C, MDA) and the sample for aroma analysis; in total 3 tubes per fruit. In brief, for each orchard,
the number of falcon tubes are: 3 (tube) x (1+2x7) (number of sampling point) x 4 (replicate) = 180
tubes. The number of tubes for 7 orchards are: 180x7 = 1260 tubes

Second, at the orchard in Hung Yen, we harvest at 4 timepoint during the dragon fruit season, to study
the effect of harvest time during the season on chilling injury. At each timepoint, we harvest dragon fruit
at 3 different maturity stages, to study the effect of maturity on chilling injury. So, in total, at Hung Yen
we harvest 12 batches of fruit. For each batch, we store the fruit similar to the fruit coming from each
orchard (described above). So actually, the orchard at Hung Yen can be considered as 12 orchards.
Therefore, on top of the above tubes, we still need to add 11 orchards, giving us an extra 180x11 = 1980
tubes

Third, at 3 of the 7 orchards, we harvest an extra batch of fruit during the off-season period. So, we have
another extra 3x180 = 540 tubes

In total, we have 1260 + 1980 + 540 = 3780 tubes


I’ve been thinking but was not sure how to reduce the number of samples.

About Senne’s work before the samples come, maybe he can first spend time working with OptiPa in
order to estimate different parameters of the respiration model, hopefully he can come up with a
proper relation between VmO2 and temperature (with a maximum).

An additional thing he can do is to do an experiment to confirm the small influence of CO2 on


respiration of dragon fruit. The result from my previous respiration showed that CO2 has significant but
small uncompetitive inhibition on respiration. This behaviour is quite different than many previous
studies that I cited in my manuscript, where a higher inhibition was observed. Although this could be
explained by the fact that in these studies, there is a confounding effect caused by the inherent inverse
correlation between O2 and CO2 concentration, it would be more convincing to confirm this with a
separate experiment with traditional respiration measurement.

About the respiration manuscript, I am still editing it and will send it next week. I am very sorry for the
delay.
