Question 1:
What is the pH of a solution of 100 mL of 0.01 M H3PO4 and
100 mL of 0.01 M Na3PO4?
Solution 1:
The two reagents will react to give a mixture compose of 0.01 mol NaH2PO4 (weak
acid) and 0.01 mol Na2HPO4 (conjugate base).
Using Henderson‐Hasselbalch equation,
pH=pKa + log [A‐]/[HA]
pH=pKa + log 0.01 mol/0.01 mol
pH=pKa + log 1
pH=pKa + 0
pH=pKa

Question 2:
Calculate the pH of a mixture of 0.25 M acetic acid and 0.10 M
sodium acetate. The pKa of acetic acid is 4.76.
Solution 2:
pH=pKa + log [A‐]/[HA]
pH=pKa + log [acetate]/[acetic acid]
pH=4.76 + log 0.10/0.25
pH=4.76 + log (0.4)
pH= 4.76 ‐0.40
pH= 4.36

Question 3:
Calculate the pH of a mixture of 0.10 M acetic acid and 0.25 M
sodium acetate. The pKa of acetic acid is 4.76.
Solution 3:
pH=pKa + log [A‐]/[HA]
pH=pKa + log [acetate]/[acetic acid]
pH=4.76 + log 0.25/0.10
pH=4.76 + log (2.5)
pH=4.76 +0.40
pH= 5.16

Question 4:
What the value of [S], as a fraction of Km, is required to obtain
80% of Vmax?
Solution 4:
Use equation for Lineweaver‐Burk Plot
Vo=80% of Vmax
Assume Vmax=1
Vo=0.8
1/Vo=Km/Vmax x 1/[S] + 1/Vmax >>> y=mx+b
1/0.8=Km/1 x 1/[S] + 1/1
1.25‐1=Km/[S]
0.25=Km/[S]
[S]/Km= 1/0.25=4
