Procedure:
2. Remove the leaf color by boiling it in alcohol. To do this, follow the steps
below:
a) Fill the beaker (3/4 of its capacity) with water. Let it boil. See Figure 3 for the
sample setup.
b) While waiting for the water to boil, get a leaf sample and place it in a test tube.
Pour denatured alcohol into the test tube until the leaf has been submerged. See
Figure 3 for the sample setup.
c) When the water in the beaker boils, place the test tube with leaf and alcohol
in it. Let the water boil for another three minutes or until all the color of the leaf
has been extracted. See Figure 4 for the sample setup.
Remove the test tube from the beaker. Then, get the leaf out of the test tube.
3. Rinse the leaf with water, and then place it on a petri dish.
If the color of the leaf turns to bluish black, it indicates the presence of starch. No
change in color indicates absence of starch.
The Mayana leaf in the test tube undergoes a rapid color shift from violet to green
when immersed in boiling water. The denatured alcohol helps in extracting
chlorophyll and therefore, helps in changing the violet color of the Mayana leaf to
color green. Due to the denatured alcohol content, it boils rapidly as well. Our
teacher added a few drops of iodine and the leaf became a bluish-black color,
indicating the presence of starch. This demonstrates that the leaf has been
performing photosynthesis and producing starch.
