DNA Quantification:

This was an automated process by the technology Nano drop spectrophotometer


that measures the concentration of extracted DNA at an absorbance of 320nm by
setting a blank with TE buffer. After extraction of DNA, it was important to know
the exact concentration and absorbance for further procedure and technique like
restriction enzyme digestion or PCR. The concentration was necessary for
determining the amounts of DNA which might be important information required
during PCR that helped to distinguished the amounts of other compounds like
primers, water, single nucleotide etc needed to be added. Nucleic acid has
heterocyclic rings that cause the maximum absorbance of ultra violet light at
260nm. In Nanodrop spectrophotometers, two ratios of absorbance are measured,
260/280nm and 260/230nm computing different significance result. 260/280nm
ratio determined the purity of nucleic acid within a range ~1.8 which also indicate
the protein contamination of the DNA, whereas 260/230nm ratio absorbance
implied the present of other organic compounds, alcohol or phenol in the DNA
with a higher absorbance value than 260/280nm absorbance value.

Table 1: The table shows 26 DNA extracted samples that were quantified out of 98
samples. Most of the concentrations and both the ratios of absorbance values were
within the expected range. R and P was the letter of the places of collected blood
samples.
