1. STAPHYLOCOCCI
gram stain : +ve ( violet coloured )
Spherical(cocci) in shape : 0.5-1
μm in diameter
Arranged in clusters (grape-like),
which is due to cell division in 3
planes and daughter cells being in
close proximity
non-motile, non-sporing
2. STREPTOCOCCI
gram stain +ve
spherical in shape
(individual cell)
arranged in pairs
and chains, which is
due to cell division
occurring in 1 plane
and daughter cells
failing to separate
completely
3. GONOCOCCI
gram stain –ve (pink
coloured)
arranged in pairs -
diplococci
cocci are kidney in shape
the line joining cocci in
pairs is perpendicular to
the long axis of cocci
intra cellular ( eg. Acute
gonorrhea – urethral
discharge )
4. CLOSTRIDIUM TETANI
gram stain +ve
drum-stick appearance
terminal and round spores-
present
5. MYCOBACTERIUM TUBERCULOSIS
acid fast bacilli – ZN STAIN/AFB
STAIN
slender, straight or slightly curved
bacilli with beaded appearance
non-sporing, non-capsulated and
non-motile
they're stained pink, the other cells
are blue ( blue background)
6. CORYNEBACTERIUM DIPHTHERIAE
albert stain is used (others:
ponders and neisser’s stain)
slender rods
arranged in angles to each
other : L, V forms , Chinese
letter pattern / cuneiform
arrangement
have polymetaphosphate/
metachromatic/ volutin/
babes earnest granules
(bluish purple) at the end
green colour bacilli
Media : pleomorphic, non-sporing,
non-capsulated
7. GRAM NEGATIVE BACTERIA
gram –ve
rod-shaped bacilli are pink
in colour
arranged individually -
scattered
eg. E.coli , proteus ,
klebsiella
8. CANDIDA SP.
gram stain +ve
oval budding yeast cells with
pseudohyphae
opportunistic fungi
causative agent for oral thrush and
vaginitis
9. CRYPTOCOCCUS
negative staining : modified india
ink stain and nigrosin stain
demonstrate the capsule
gram stain +ve round budding
yeast cells
causative agent for meningitis
can be grown on SDA (Sabouraud
dextrose agar)
