II. RATIONALE

CHED Memorandum Order No. 23, S. 2021, mandates that State Universities

and Colleges (SUCs) offering agriculture programs must rigorously adhere to the

program's minimum requirements. This includes full exposure of students to various

platforms of practical learning in agriculture to allow students to gain practical, real-world

experience that reinforces classroom theory and deepens their comprehension of crop

science. Moreover, it fosters skill development by equipping students with critical agricultural

skills such as plant propagation, soil analysis, pest management, and data collection, which

directly prepare them for agricultural careers. These laboratory activities also cultivate

problem-solving abilities, vital for addressing real-world challenges in agriculture.

Specialized Propagation Techniques is an educational program designed to

provide students with a unique opportunity to explore the fascinating world of plant
tissue culture and its pivotal role in enhancing crop production. In this activity,

students will delve into the science and practice of tissue culture, learning how this

advanced agricultural technique can contribute to crop improvement, disease

resistance, and genetic preservation. Through a combination of theoretical

knowledge and hands-on laboratory experiences, students will gain insights into the

vital role that tissue culture plays in modern agriculture, enabling them to appreciate

its significance in addressing global food security challenges. Exposing them into

tissue culture activities will not only broaden their understanding of plant biology but

also contribute to the advancement of sustainable agricultural practices.

III. OBJECTIVES:

1. Introduce the concept of tissue culture to students and an understanding of

tissue culture as a specialized propagation technique and its significance in

modern agriculture.

2. Hands-on experience in setting up tissue cultures and practical exposure to

the aseptic techniques and procedures involved in tissue culture, allowing

them to develop essential laboratory skills.

3. Educate students on the diverse applications of tissue culture in crop

improvement, including micropropagation, disease resistance breeding, and

genetic modification.

IV. METHODOLOGY

The students will be oriented first with the basic tissue culture activities where

they would learn the aseptic techniques, proper handling of laboratory equipments,

maintaining sterile conditions as well as the safety guidelines and rules in the

laboratory.
Hands-on Laboratory Activities:

a. Collection and Sterilization of Banana Suckers

Collected banana suckers will be placed in a sterile container or bag

immediately after cutting to prevent contamination from the environment.

b. Workstation Setup

The students will be divided into small groups, each with its own workstation

equipped with sterile equipment and materials.

c. Plant Tissue Preparation

Students will be instructed on how to prepare and sterilize plant tissue

samples (explants) for culture.

d. Culture Media Preparation

Demonstrate to them the preparation of sterile culture media, explaining its

composition and purpose.

e. Inoculation and Incubation


Guide students through the process of inoculating the explants into the culture

vessels and setting up cultures under sterile conditions. Explain temperature

and light requirements for different plant species.

f. Microscopy

Provide students with the opportunity to examine the cultures under

microscopes to observe cell growth and development.

g. Documentation

Encourage students to keep records of their experiments, including

observations and results

h. Assessment
Students' understanding and skills will be assessed based on their

participation, successful completion of hands-on tasks, and their ability to

engage in the discussion and present their ideas.
