DILUTION PROBLEMS

i. How would you prepare:


a. 10 mL of a 1:10 dilution of a 1 M NaCl solution and what would the final
concentration of NaCl?
1 ml of 1M NaCl with 9 ml of water mix together. The final concentration for NaCl is 0.1M.

b. 80 mL of a 1:20 dilution of a 1 M NaCl solution?


4 ml of 1M NaCl mix together with 76 ml of water. The final concentration of NaCl is
0.05M.

C. 50 mL of a 1:25 dilution of a 1 M NaCl solution?


2 ml of 1M NaCl mix with 48 ml of water. The final concentration of NaCl is 0.04M.

ii. How would you prepare exactly 6 mL of a 1/20 dilution (assume the concentration of
your
starting solution is “1”)?
0.3 ml of stock mix with 5.7 ml of water. The final concentration of NaCl is 0.05M.

iii. You are provided with an antibody solution (Ab) that has a concentration of 600
microgram (μg) / microlitre (μL). For lab, it is necessary to make the following
dilutions:
a. 10 μL of 600 μg/μL Ab + 190 μL of buffer to make a 1:20 dilution at X μg/ μL.
M1 = 600μg/ μL
V1 = 10μL
V2 = (190+ 10)
= 200 μL
M1V1 = M2V2
(600 μg/μL)(10 μL) = (M2)(200 μL)
M2(X) = 30 μg/μL
b. 20μL of 1:20 Ab + 40μL of buffer to make a 1:60 dilution at X μg/μL.
M1 = 1:20 x 600 μg/μL
= 30 μg/μL
V1 = 20μL
V2 = (40+20)
= 60μg/μl
M1V1 = M2V2
(30 μg/μL)(20 μL) = (M2)(60 μg/μL)
M2(X) = 10 μg/μL

c. 5 μL of 1:60 Ab + 5 μL of buffer to make a X dilution at Y μg/μL.


M1 = 1:60 x 600 μg/μL
= 10 μg/μL
V1 = 5 μL
V2 = (5+5)
= 10 μL
M1V1 = M2V2
(10 μg/μL)(5 μL) = (M2)(10 μL)
M2(Y) = 5 μg/μL
1:X x 600 μg/μL = 5 μg/μL
600/X = 5 μg/μL
X = 120
X = 1:120
d. 10 μL of 1:60 Ab + 90 μL of buffer to make a X dilution at Y μg/μL.
M1 = 1:60 x 600 μg/μL
= 10 μg/μL
V1 = 10 μL
V2 = (90+10)
= 100 μL
M1V1 = M2V2
(10 μg/μL)(10 μL) = (M2)(100 μL)
M2(Y) = 1 μg/μL
1:X x 600 μg/μL = 1 μg/μL
600(X) = 1 μg/μL
X = 600
X = 1:600

e. 10 μL of 1:60 Ab + 40 μL of buffer to make a X dilution at Y μg/μL.


M1 = 1:60 x 600 μg/μL
= 10 μg/μL
V1 = 10 μL
V2 = (40+10)
= 50 μL
M1V1 = M2V2
(10 μg/μL)(10 μL) = (M2)(50 μL)
M2 (Y) = 2 μg/μL
1:X x 600 = 2 μg/μL
X = 300
X = 1:300
f. 10 μg of 1:60 Ab + 10 μL of buffer to make a X dilution of Y μg/μL.
M1 = 1:60 X 600 μg/μL
= 10 μg/μL
V1 = 10 μL
V2 = (10+10)
=20 μL
M1V1 = M2V2
(10 μg/μL)(10 μL) = (M2)(20 μL)
M2 (Y) = 5 μg/μL
1:Xx600 μg/μL = 5 μg/μL
600/X = 5 μg/μL
X = 120
X = 1:120

iv. How much 2.0M NaCl solution would you need to make 250 mL of 0.15 NaCl
solution?
M1 = 2.0M
M2 = 0.15 M
V2 = 250 mL

(2.0 M)(V1) = (0.15 M)(250 mL)
V1 = 18.75 mL
v. What would be the concentration of a solution made by diluting 45.0 mL of 4.2 M
KOH to 250 mL?
M1 = 4.2 M
V1 = 45.0 mL
V2 = 250 mL
M1V1 = M2V2
(4.2 M)(45.0 mL ) = (M2)(250 mL )
M2 = 0.756 M

vi. What would be the concentration of a solution made by adding 250 mL of water to
45.0 mL of 4.2 M KOH?
M1 = 4.2 M
V1 = 45.0 mL
V2 = 295 mL
M1V1 = M2V2
(4.2)(45.0) = (M2)(295)
M2 = 0.641 M

vii. How much 0.20 M glucose solution can be made from 50.0 mL of 0.50 M glucose
solution?

(0.50 M)(50.0 mL) = (0.20 M)(V2)
V2 = 125 mL
viii. What is the molarity of a solution that has 4.5 mol of solute dissolved in 300 ml of
solution?
