Title: Denaturation of Proteins Through Coagulation
I. Learning Objectives
To Identify what is denaturation.
Determine how denaturation works.
II. Materials
A. Chemicals/Reagents
Alcohol
Egg
Salt
B. Glasswares
Plastic Spoons
Styrofoam Bowls
III. Procedure
1.) Prepare the materials.
2.) Separate the egg whites from the egg yolk. Set aside the yolk.
3.) Fill the Styrofoam bowls with egg whites and label them.
4.) Add 1 teaspoon of salt on one bowl and mix. Then add another
teaspoon and mix it again and observe.
5.) Then on the second bowl, add 1 teaspoon of isopropyl alcohol to the
egg white. Stir and observe.
IV. Results and Observation
The proteins that are present in the albumin started to react and
coagulate when mixed with salt and alcohol. As proteins deform or
unravel parts of structure that were hidden away get exposed and form
bonds with other protein molecules, so they coagulate (stick together)
and become insoluble in water. Salt is capable of disrupting bonds and
interaction in molecules thus leading to the change of protein structure
denaturing them. And for the alcohol egg white, the alcohol is a form of
natural solvent, these solvents are capable altering the protein structure
through the formation of ne hydrogen bonds that are found between the
alcohol and protein side chains.
