IV. PROCEDURES ELICIT

A. Reviewing previous THUMBS UP or THUMBS DOWN . The teacher will ask the students to

1. When observing a specimen under the microscope it is important to start

viewing from a lower power objective to high power objectives.
2. Observing a specimen is still possible even if the diaphragm is closed.
3. You need to look at the microscope from the side when turning the coarse
adjustment knob to lower the objective lens to avoid breaking the lens.
4. The specimen on the slide should be placed right above the hole of the
stage.
5. Turn the coarse adjustment knob to get a sharper image.

1. The class will be divided into five groups.
2. Each group will be provided with an activity sheet with the activity
procedures to follow. (See attached Activity Sheet)

1. Why must a specimen needs to be very thin to be viewed under the
microscope?
2. What will happen if wet mounting of a specimen was not done
properly?
EVALUATION
Directions: Arrange the steps in correct order when wet mounting a
specimen. Number your answers from 1-5 and write it on the blank before
the number.
____1. Position the cover slip at 45 degrees with one side touching one edge of the
water on the slide.
____2. Add a drop of water over the specimen.
____3. Slowly lower the other edge of the cover slip until it rests on the
water and the printed letter.
____4. Remove or minimize trapped bubbles by gently tapping the cover slip.
____5 . Place the specimen on the center of a glass slide.

Activity Sheet
Making a Wet Mount
Materials needed:
Slide Scissors
Cover Slip Water
Dropper Specimen: Letters e, n, m, r,
Tweezers
Procedures:
1. Cut out a small letter “e” from a newspaper page using a scissor.
2 . Using tweezers, place the small letter “e” in the center of a glass slide in an upright
position.
3. Using a dropper, add 1 drop of water over the specimen. It will act as a mounting
medium and make the image of the specimen clear.
4. Get the coverslip and position the cover slip at 45 degrees with one side touching one
edge of the water on the slide. Slowly lower the other edge of the cover slip until it rests
on the water and the printed letter. Bubbles are perfect circles you see on your preparation.
Remove or minimize trapped bubbles by gently tapping the cover slip with the eraser-end of a
pencil. Make the bubble move towards the edge of the cover slip.

5. If you’re done, present your prepared slide to your teacher.

6. Create an illustration of the procedures you did.
