Deoxyribonucleic Acid
DNA is the nucleic acid found in the nucleoid of prokaryotes and the
nucleus of eukaryotes.
สารพันธุกรรมหลักที่พบในสิ่งมีชีวิตเกือบทุกชนิด ยกเว้นไวรัสบางกลุ่ม คือ DNA ซึ่ง
เป็นกรดนิวคลีอิก (nucleic acid) ที่พบในบริเวณนิวคลีออยด์ของเซลล์โพรคาริโอต
และในนิวเคลียสของเซลล์ยูคาริโอต
Because DNA is made up of many phosphate molecules, it contains
a negative ion. As a result, the curling of the DNA is restricted. Histone
protein, which contains arginine and lysine, helps Eukaryotic cells handle
this problem.

DNA REPLICATION
Helicase
Helicases are enzymes that untwist the double helix at the replication forks
• Single-strand binding proteins bind to and stabilize single-stranded DNA
• Topoisomerase corrects “overwinding” ahead of replication forks by
breaking, swiveling, and rejoining DNA strands

DNA Polymerase
DNA synthesis is actually a short stretch of RNA, not DNA. This RNA chain is
called a primer and is synthesized by the enzyme primase.

DNA polymerases cannot initiate synthesis of a polynucleotide; they can only add
nucleotides to the 3′ end
• The initial nucleotide strand is a short RNA primer.
• An enzyme called primase can start an RNA chain from scratch and adds RNA
nucleotides one at a time using the parental DNA as a template.

DNA Replication
DNA Polymerase
• The primer is short (5–10nucleotideslong),and the 3′ end serves as the
starting point for the new DNA strand.
• Enzymes called DNA polymerase 3 catalyze the elongation of new DNA at
a replication fork.
DNA Ligase
DNA ligase serves as the welding of the incoming DNA lines, replacing the
primary RNA filled with the previously synthesized DNA lines. It also serves
to weld the Okazaki fragment as the next lagging strand.
