Synapse
1. Axodendritic Synapse :
เมือมีการจับของ Neurotransmitter
กับ receptors จะมีการส่ งของสั ญญาณ
ต่ อเข้ าไปใน cells และทําให้ มีการเปลียนแปลง
ภายใน neuron receptors ทีเป็ นตัวรับแบ่ ง
ได้ เป็ น 2 class
Ligand – gated channels :
receptors ชนิดนีจะมีการเปิ ดของ
channel เมือมีการจับของ
neurotransmitter หรื อ ligand
แล้วทําให้ มปี ระจุผ่าน channel เข้ าใน
cells ถ้ าเป็ นประจุบวกผ่ านเรียก
“excitatory” ถ้ าเป็ นกลุ่มทีทําให้ ประจุลบ
ผ่ านเรียก “inhibitory”
- major excitatory
Neurotransmitter ได้ แก่
Glutamate
- major Inhibitory
Neurotransmitter ได้แก่
GABA
II. G-Protein linked receptors : การ
จับของ neurotransmitter ที receptor
จะกระต้ นุ ให้ intracellular proteins ที
เรียกว่ า “G-Proteins” (ที
เรียกชื อนีเพราะว่ ามีการจับกับ
Guanine nucleotide)
Neurotransmitter และ
neuromodurator การกระตุ้นต่ อ
receptors ได้ โดยมีผลต่ อ PI turnover,
การเปลียนแปลงของ ion influx หรื อ
inhibition ต่ อ cAMP อาจเกิดหนึงหรื อ
มากกว่าหนึงปฏิกริ ิยาก็ได้
โดยส่ วนใหญ่ การเกิดของสั ญญาณเริม
จากการมี depolarization ของ
membrane ในส่ วนของ Receptor ทีมี
ความเกียวข้ องกับ G Protein (หรื อ
Guanine nucleotide-binding protein)
Receptor ทีใช้ pathway นีได้แก่
1. Adrenergic receptors ทังหมด
2. 13 ถึง 14 serotonin receptors ทีรู้จักอยู่ในขณะนี
3. Dopamine receptors ทัง 5 ชนิด (ทีรู้จักกันในขณะนี)
4. Muscarinic receptors ทัง 5 ชนิด (ทีรู้จักกันในขณะนี)
5. Neuropeptide receptors ทังหมด
6. บางส่ วนของ Glutamate และ GABA receptors จะ
เป็ นทัง ligand gated channel และ G-protein
linked
G-Protein มีหลาย Types :
1. C-AMP Production
2. P I Turnover (PI = Phosphoinositide)
3. Effect ของ G-Protein ต่ อ ion channel (ปฏิกริ ิยานีจะ
น้ อยกว่าทีเกิดจาก Ligand gated channel โดยตรง)
การ Activate G-Protein
เมือมี Neurotransmitter มาจับกับ receptor ทําให้ มี depolarization
และทําให้ มีการเปลียนแปลงใน receptor ทีเกียวข้ องกับ G-Protein ซึงมี
ผลให้ MG++ ไปจับกับ GTP (Guanosine 5’-triphosphate) ทําให้ เกิด
พลังงานไปสลาย Trimeric G-Protein ให้ เป็ น component ของ alpha
beta และ gamma subunits สั ญญาณทีซับซ้ อนทีเกิดขึนจะขึนอยู่กับ
ชนิดของ Neurotransmitter ทีกระตุ้นต่ อ G-Protein และทําให้ มี
ปฏิกิริยากับ second messenger ทีแตกต่างออกไป
Receptor linkage to intracellular effector cnzymes to catalyze second
messenger formation. G=guanine nucieotied-binding protien ; GDP =
guanosine B’-dlphosphate; GTP = gusnosine 5’ – triphosphate; cAMP =
sdenosine 3, 5’-cycle monophosphate; Pl = phosphoinositide.
สาร G-like proteins สามารถกระตุ้น
phospholipase C ซึง Phopholipase C ทํา
ให้ มี Hydrolyze ของ Phosphatidy-linosital
4,5 – biphosphate และทําให้ เกิดสาร
Diacyglycerol และ inositol 1,4,5-
triphosphate ซึงมีผล oderate การทํางานของ ion
channel ได้
พบ Gi-Protein ตัวอืน ให้ ชือว่า Go (o=other protein
ซึงก็ยังไม่ พบว่ ามี function อะไรทีชัดเจน
ความรู้เท่าทีรู้ในปัจจุบัน พบว่ า G Proteins ทีมีความเกียวข้ องกับ
Signal transduction ได้ แก่ α γ
heterotrimers
การกระตุ้นต่ อ G Protein ก็มีผลกระตุ้นต่ อ Catalytic
subunit ของ membrane bound adenyl cyclase หรื อ
ในกรณีทมีี การลดการกระตุ้นของ G protein ก็ให้ ผล
ตรงข้าม ขบวนการทีเกิดขึนในเซลส์ เกิดขึนโดยใช้
phosphorylating enzyme เป็ นสื อกลาง เช่ น ใช้
phosphokinase A (PKA) เป็ นต้ น
Signal transduction ผ่าน receptors
เฉพาะชนิดมีบทบาทสํ าคัญในการกระตุ้นสัญญาณประสาท
การศึกษาพบว่ า G-Protein มีความเกียวข้ อง โดยทํา
หน้ าทีและบทบาทในการขยายหรื อลดสัญญาณทีส่ งผ่ านมาจาก
ภายนอกเซลส์ และส่ งต่ อในภาพรวมเป็ นสั ญญาณทีกลันกรอง
แล้วไปสู่ effectors ปริมาณของ G protein ไม่
คงที แต่ เปลียนไปตามการเปลียนแปลงทางสรีรวิทยา, พยาธิ
สภาพ และการก่อกวนจากยา ได้ มีการพบความผิดปกติของ G
protein function ในโรคทางอายุรกรรมและโรค
สําคัญทางจิตเวชหลายโรค
