뇌신경계 질환은 바이러스 벡터를 이용한 치료 방법을 이용하고 있으며 glutamic acid
decarboxylase (GAD) 혹은 aromatic l-amino acid decarboxylase (AADC) 유전자를 아데노 부속
바이러스에 삽입하여 치료 효능을 분석하고 있다. 유전자 치료제는 세부 질환에 따라 그 주입
방식이 다르며 뇌질환(파킨슨병 및 알츠하이머)은 뇌 조직에 직접 투여하며 암으로 인한 국소통증
해당 부위의 진피에 주입하는 방법을 이용한다. 뇌신경계 질환 유전자 치료제는 약리작용을 가지는
치료 단백질을 발현하여 분비하는 작용기전을 나타내는 것으로 알려져 있다.
대표적으로 파킨슨병의 경우 아데노 부속 바이러스 벡터에 글루타민산탈탄산 효소를
삽입한(AAV-GAD) 임상 시험 결과, 유해성이 나타나지 않았으며 환자의 운동성이 개선됨을
확인하였다. 또한 신경영양인자(glial cell line derived neurotrophic factor) 대체 뉴트린(NTN)를
아데노 부속 바이러스 벡터에 삽입 후 치료제의 안정성과 내약성을 검증한 결과 부작용이 확인되지
않았다.

2.3.5 감염질환
현재 유전자 치료제의 10분의 1가량은 다국가 임상 연구가 진행되고 있다. 국가별 임상연구
현황은 미국(53%), 영국(6%), 프랑스(5%), 중국(3%), 한국(2%) 순으로 나타났다. 절반 이상의 임상
연구가 진행중인 미국의 경우 암과 유전질환의 연구가 가장 많이 수행되고 있다. 우리나라는
진행되고 있는 11건의 임상연구 중 조직별 질환(5건), 유전질환(2건), 감염질환(2건) 및 암(1건)이
진행되고 있다[8]. 유전자 치료제 개발 국가의 대다수가 난치성 질환을 대상으로 연구를 수행하고
있으나 우리나라는 근골격계 질환 관련 연구가 주로 수행되고 있는 특징이 있다.
