Operon gồm:
• Nhóm gen cấu trúc: bao gồm các gen nằm kề
nhau mã hóa cho các enzyme, protein có liên
quan với nhau về mặt chức năng.
• Vùng vận hành (Operator): nằm trước nhóm gen
cấu trúc, chỉ huy sự hoạt động của nhóm gen cấu
trúc.
• Vùng khởi động (Promoter): nằm trước vùng vận
hành, là nơi để RNA polymerase bám vào và
phiên mã.

• Còn có gen điều hòa (Regulator), không thuộc
operon, nằm liền kề hoặc cách xa operon.
• Gen điều hòa Phiên mã Dịch mã Protein
ức chế (repressor) tương tác với Operator
Điều chỉnh hoạt động của nhóm gen cấu trúc:
- Repressor bám vào Operator -> không phiên mã.
- Repressor không bám vào Operator -> phiên mã.

Repressor có thể ở dạng hoạt động hoặc bất hoạt
tùy vào sự hiện diện của các phân tử nhỏ khác:
Inducer (chất cảm ứng) là phân tử có khả năng kết
hợp với repressor, bất hoạt repressor và cảm ứng
“mở” operon.
Corepressor (chất đồng ức chế) là phân tử có khả
năng kết hợp với repressor, hoạt hóa repressor và ức
chế (“tắt”) operon.

• Lactose: đóng vai trò là chất cảm ứng (inducer) giúp mở operon
lac.
• Một operon cảm ứng là dạng bình thường
“tắt”; một phân tử chất cảm ứng (inducer) sẽ
bất hoạt repressor và giúp xảy ra phiên mã
gen cấu trúc.
• Các enzyme được điều hòa theo kiểu cảm
ứng thường có chức năng trong các con
đường dị hóa; sự tổng hợp chúng được cảm
ứng bởi tín hiệu hóa học.

6.2.3. Điều hòa sau phiên mã
• Sự phiên mã chưa bảo đảm là gene sẽ được biểu
hiện.
• Các cơ chế điều hòa có thể hoạt động ở nhiều giai
đoạn sau phiên mã, cho phép TB điều chỉnh sự
biểu hiện gene trong đáp ứng với sự thay đổi môi
trường tốt hơn.
a. Sự chế biến, cải biên RNA
• Cắt nối thay thế (alternative RNA splicing): ghép
nối các exon theo các cách khác nhau tạo ra các
loại mRNA trưởng thành khác nhau từ 1 loại tiền
mRNA.
b. Sự thoái biến mRNA
• Thời gian tồn tại của mRNA trong TBC là chìa khóa
xác định lượng protein được tổng hợp.
• mRNA của eukaryote tồn tại lâu hơn mRNA
prokaryote.
• Thời gian tồn tại của mRNA được xác định chủ yếu
bởi các vùng không được dịch mã ở đầu 5’ và 3’
của mRNA.
6.2.4. Điều hòa dịch mã
• Sự khởi đầu dịch mã có thể bị chặn lại bởi các
protein điều hòa khi chúng gắn vào mRNA.
• Sự dịch mã của tất cả mRNA trong tế bào có thể
được điều hòa đồng thời.
– Chẳng hạn, các nhân tố khởi đầu dịch mã được hoạt hóa
cùng lúc trong tế bào trứng sau khi thụ tinh.
6.2.5. Điều hòa sau dịch mã
• Sau khi dịch mã, protein được biến đổi theo nhiều
kiểu khác nhau, bao gồm cả sự cắt và thêm các gốc
hóa học, các quá trình này cũng có thể được kiểm
soát.
• Proteasome là những phức hợp protein lớn liên kết
với các phân tử protein và phân hủy chúng.

Pattern Formation: Setting Up the Body
Plan
• Pattern formation is the development of a spatial
organization of tissues and organs
• In animals, pattern formation begins with the
establishment of the major axes
• Positional information, the molecular cues that
control pattern formation, tells a cell its location
relative to the body axes and to neighboring cells
• Pattern formation has been extensively studied in
the fruit fly Drosophila melanogaster
• Combining anatomical, genetic, and biochemical
approaches, researchers have discovered
developmental principles common to many other
species, including humans

• This phenotype suggests that the product of the
mother’s bicoid gene is concentrated at the future
anterior end
• This hypothesis is an example of the gradient
hypothesis, in which gradients of substances called
morphogens establish an embryo’s axes and other
features
• The bicoid research is important for three reasons:
– It identified a specific protein required for some
early steps in pattern formation
– It increased understanding of the mother’s role in
embryo development
– It demonstrated a key developmental principle that
a gradient of molecules can determine polarity and
position in the embryo
