从文献和化学分析角度探讨附子的用量与煎服方法

摘要

目的 :对古今临床附子使用量和煎服方法进行整理’并从化学分析方面探讨在

不同 附子用量下’煎煮后生物碱成分的变化’以及与甘草配伍煎煮后生物碱成

分的变化。

方法 :对〈伤寒杂病论〉、“火神派”代表性中医人物以及文献报道的有共同才子

雯用 情况进行总结归纳和分析’根据整理的应用经验制定 7 组附子黑顺片单煎

工样品和 6 组附子甘草合煎液样品’进行 3 0 分钟的预浸泡和 2 小时的文火煎

安,然后浓缩水煎液并定容到 lOOml 容量瓶中, 浓缩过程产生的固体再用甲醇

兰行抽提,并定容到 lOOml 容量瓶’每组水煎j夜样品稀释 100 倍后进样,沉淀

三三享提取液直接进样进行 UPLC-MS 分析’分析样品中的生物碱成分并做组间比

三果 : 附子单煎液组别中每克附子的双醋型生物碱煎出量随同才子用量增大而减

少 ,而附子甘草合煎液组别中每克附子的双醋型生物碱煎出量受甘草所占比例

一← , I、 的影响 ;二次乌头碱为所有组别的主要检出的生物碱成分,在 lOOg 附子单煎

主专 30 g 附子甘草合煎液中每克附子煎出量的峰值’分别为 233320ng 和

! : ~560口g 。

子用量的增大而减慢;甘草所占比例越大’双目旨型生物碱煎出量越少;甲醇比

超纯水可能更加适合用于提取附子所含的各种生物碱成分 。
