The solubility of proteins is strongly dependent on the salt concentration (ionic strength)
of the medium. Proteins are usually poorly soluble in pure water. Their solubility increases
as the ionic strength of certain ions increases, because more and more of the well-
hydrated inorganic ions (blue circles) are bound to the protein’s surface, preventing
aggregation of the molecules (salting in). At very high ionic strengths, the salt withdraws
the hydrate water from the proteins and thus leads to aggregation and precipitation of the
molecules (salting out). For this reason, adding salts such as ammonium sulfate
(NH4)2SO4 makes it possible to separate proteins from a mixture according to their degree
of solubility (fractionation).

➢Како ќе ја зголемиме брзината на растварање?
➢Tемпературата е значаен фактор за определувањето на растворливоста на
лековитата супстанца и за подготовката на раствор од истата.
➢При растварање на повеќето хемиски супстанции апсорбирањето на топлината од
околината резултира со зголемување на растворливоста (ендотермен процес), при
што со загревање со подобрува растворливоста. Кај мал број хемиски супстанции
растворливоста се намалува со зголемување на температурата.
➢Врз растворливоста освен температурата влијание може да имаат и други
фактори како што се различните хемиски и други физички карактеристики на
растворената супстанција и на растворувачот, притисок, pH на растворот, степенот
на иситнетост на супстанцијата, и употребата на мешање при процесот на
растварање.
➢Растворливоста на чиста хемиска супстанца на дадена температура и притисок е
константна; сепак степенот на растварање, кој претставува брзината со која таа се
раствора, зависи од големината на честичките на таа супстанција и од степенот на

Practically speaking, this is a fairly simple problem to solve. Solutions are prepared
containing varying concentrations of ethanol or acetone in water, ranging from 0 to
100%. The required concentration of drug is added to each solution and the solutions
are refrigerated overnight, then viewed for precipitation.
In our example, we see that at least 60% v/v ethanol is required to solubilize our drug.
This leaves us with a problem since, from a flavoring standpoint, 60% ethanol is not
considered pharmaceutically elegant. (Why?) We can now take this information and
determine the dielectric constant of the solvent system which provides drug solubility.

Sandimmune® Soft Gelatin Capsules (cyclosporine capsules, USP) and
Sandimmune® Oral Solution (cyclosporine oral solution, USP) have
decreased bioavailability in comparison to Neoral® Soft Gelatin
Capsules (cyclosporine capsules, USP) MODIFIED and Neoral® Oral
Solution (cyclosporine oral solution, USP) MODIFIED. Sandimmune®
and Neoral® are not bioequivalent and cannot be used interchangeably
without physician supervision. The absorption of cyclosporine during
chronic administration of Sandimmune® Soft Gelatin Capsules and Oral
Solution was found to be erratic. It is recommended that patients
taking the soft gelatin capsules or oral solution over a period of time be
monitored at repeated intervals for cyclosporine blood concentrations
and subsequent dose adjustments be made in order to avoid toxicity
due to high concentrations and possible organ rejection due to low
absorption of cyclosporine. This is of special importance in liver
transplants. Numerous assays are being developed to measure blood
concentrations of cyclosporine. Comparison of concentrations in
published literature to patient concentrations using current assays
must be done with detailed knowledge of the assay methods
employed. 110

trough concentration, cyclosporine exposure will be greater with

arthritis patients taking Neoral® to avoid toxicity due to high

DEIONIZATION

Deionization is a physical process which uses specially-
manufactured ion exchange resins which bind to and
filter out the mineral salts from water.
Because the majority of water impurities are dissolved
salts, deionization produces a high purity water that is
generally similar to distilled water, and this process is
quick and without scale buildup.
However, deionization does not significantly remove
uncharged organic molecules, viruses or bacteria,
except by incidental trapping in the resin.
Specially made strong base anion resins can remove

THERMO-COMPRESSION DISTILLATION
The water is heated to a temperature slightly above 100°C and is then as steam led to a
compressor. When the steam is compressed the temperature is increased and the steam is led
back to the still where it is condensed while heating the still and the feed water. As all the
evaporation energy is returned to the process, and only a limited amount of cooling water is
needed, the process is economical for production of large quantities of WFI.
The principle of thermocompression can be utilised for Multiple-Effect Distillation The steam is
generated under high pressure and has a higher temperature. This steam is used to heat the
second station working at a lower pressure. The steam is thereby condensed giving off its heat of
condensation. This cycle can be repeated for a number of stations, usually four to six.

THERMO-COMPRESSION DISTILLATION
Different contaminants like particles, ions, endotoxins and other biological
substances can be carried over in unevaporized water droplets together with
the steam. To avoid this it is essential to have efficient separation systems.
Bigger drops can effectively be removed by gravity separation when the
steam has a low velocity. Drops will return to the system as pre-heated feed
water to the next effect. The steam can also be accelerated and guided in a
tangential flow through a cyclone. The centrifugal force, in a high velocity cyclone, is
over 500 G and will force droplets bigger than approximately 1 μm towards the outer
tube wall from which they will flow down to the drain and go over as feed water
to the next effect. It is also an advantage if the steam changes direction through the
separation system. To increase the safety a combination of the different separation
systems is often used.
